Description
It contains; Amlodipine and Atorvastatin. Amlodipine: Antianginal Agent; Antihypertensive; Calcium Channel Blocker, Dihydropyridine. Amlodipine is a dihydropyridine calcium antagonist it acts by inhibiting the transmembrane influx of calcium ions into vascular smooth muscle and cardiac muscles this mechanism leads to a reduction in peripheral vascular resistance, resulting in a decrease in blood pressure. Atorvastatin is a lipid-lowering agent. Atorvastatin is an HMG Co-A reductase inhibitor, competitively inhibits 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase. This enzyme catalyzes the conversion of HMG-CoA to mevalonate, an early and rate-limiting step in cholesterol biosynthesis, this mechanism results in a compensatory increase in the expression of LDL receptors on hepatocyte membranes and a stimulates catabolism of LDL. HMG-CoA reductase inhibitors not only decrease the levels of high-sensitivity C-reactive protein (hsCRP), but they also exhibit pleiotropic properties like; improving endothelial function, reduction in inflammation at the site of the coronary plaque, inhibition of platelet aggregation, and anticoagulant effects . This drug is indicated for patients with primary hyperlipidemia and mixed dyslipidemia as an adjunct to diet to reduce elevated total-Cholesterol, LDL-C, non HDL-C, and TG levels and to increase HDL-C, in patients with hypertriglyceridemia as an adjunct to diet.

Dosage
Dose must be taken according to doctor’s advice only; doses given below are suggested as supportive literature for better understanding in some areas of requirements. Adult dose: For prevention of cardiovascular disease, hypertension/angina & hyperlipidemia; dosage range is 2.5-10mg amlodipine/10-80mg atorvastatin orally a day. Dose must be adjusted according to patient’s condition. Do not exceed up to 10mg amlodipine/80mg atorvastatin a day.
-
In case of Overdose
There is no specific treatment in the event of overdose. In the event of overdose, the patient should be treated symptomatically and supportive measures instituted as required. In overdose amlodipine may cause hypotension.
-
Missed Dose
In case you missed any dose, take the next dose as soon as you remember, but don't double your next dose to compensate for the missed one.
-
How To Use
Can be administered as a single dose at night, with or without food or as suggested. The tablet should be swallowed whole.
-
When Not to Use
Hypersensitivity to amlodipine or any component of the formulation occurs, then do not use. Do not use amlodipine during breastfeeding. Atorvastatin should not be used in patients with a known hypersensitivity to any component of this product. Atorvastatin should not be used in patients with active liver disease or with unexplained persistent elevations of serum liver enzymes, in pregnancy and breastfeeding.

Side Effects
Myalgia Constipation Asthenia Abdominal Pain Nosebleeds Nausea Peripheral edema palpitations Pruritus
-
Precautions & Warnings
In case of amlodipine; symptomatic hypotension can occur; acute hypotension upon initiation is unlikely due to the gradual onset of action. Blood pressure must be lowered at a rate appropriate for the patient's clinical condition. Use with caution in patients with hepatic impairment. Atorvastatin should be used with caution in patients who have a history of liver disease, renal impairment.

Food Interactions
Diet modification and monitoring is mandatory with use of lipid lowering agents. Grapefruit juice may increase serum concentration of atorvastatin. Red yeast rice are rich with variable quantities of different compounds that have structural similarity to HMG-CoA reductase inhibitors, primarily monacolin K (or mevinolin) which is structurally identical to lovastatin; so concomitant use of red yeast rice with HMG-CoA reductase inhibitors may result in increased incidence of adverse or toxic effects of this drug.
