It primarily inhibits bacterial protein synthesis by binding to the elongation factor G (EF-G) in susceptible bacteria. EF-G is essential for the translocation step during protein synthesis, where it facilitates the movement of the ribosome along the messenger RNA (mRNA) template. By binding to EF-G, fusidic acid prevents the release of EF-G from the ribosome, thereby blocking the translocation process. This inhibition leads to the disruption of protein synthesis, ultimately inhibiting bacterial growth and replication. Fusidic acid's selective binding to EF-G in bacteria, which is structurally different from the corresponding factor in mammalian cells, contributes to its antibacterial specificity and minimal impact on human protein synthesis.
