    Ofloxacin, a synthetic chemotherapeutic antibiotic, exerts its bactericidal effect by inhibiting DNA gyrase and topoisomerase IV, essential bacterial enzymes responsible for DNA replication, transcription, and repair. This inhibition leads to DNA fragmentation and cell death in susceptible bacteria. The primary target of ofloxacin is DNA gyrase, a type II topoisomerase, which introduces negative supercoils into DNA, a necessary step in DNA replication. By blocking the action of DNA gyrase, ofloxacin prevents the relaxation of supercoiled DNA, thereby disrupting the bacterial cell cycle and leading to cell death. Additionally, ofloxacin can also inhibit topoisomerase IV, another essential enzyme involved in DNA replication and segregation during bacterial cell division.
