  • Description

    Rabeprazole sodium, is a substituted benzimidazole that inhibits gastric secretion. Inhibition of the proton pump by rabeprazole is partially reversible. It has 2- to 10-fold greater antisecretory activity than omeprazole but it dissociates more readily from H+,K+-ATPase than omeprazole, therefore it has a shorter duration of action. Rabeprazole is an alpha-pyridyl methyl sulfinyl benzimidazole drug and a selective and irreversible proton pump inhibitor with antisecretory properties. Rabeprazole works by entering into the parietal cell and accumulating in the acidic secretory canaliculi where this drug is activated by a proton-catalyzed process and this process leads to formation of a thiophilic sulfonamide or sulfenic acid. This activated rabeprazole now forms covalent bonds with the sulfhydryl amino acids cysteine on the extracellular domain of the proton pump (H+/K+ ATPase) present at the secretory surface, this mechanism inhibits the transport of hydrogen ions, via exchange with potassium ions, into the gastric lumen. Binding of this drug to cysteine 813, in particular, is necessary for inhibition of gastric acid production.

    Treatment of symptomatic GERD, non-erosive and erosive gastro esophageal reflux diseases. Healing of gastric and duodenal ulcers. Helicobacter pylori eradication to reduce the risk of duodenal ulcer recurrence. Treatment of pathological hypersecretory conditions, including Zollinger-Ellison syndrome.
  • Dosage

    Dose must be taken according to the doctor’s advice only; doses given below are suggested as supportive literature for better understanding in some areas of requirements. Adults dose: For gastroesophageal reflux disease; 10-20 mg once daily. For H-pylori removal; 20-40mg twice daily. For Zollinger-Ellison syndrome, Starting dose; 60mg once daily, maximum dose; 100mg once daily or 60mg twice daily. Child dose: Infants: Sprinkle capsules: 5 to 10mg orally once daily. Children age ≤11 years: Sprinkle capsules: weight <15 kg; 5mg orally once daily; may increase to 10mg once daily, weight ≥15 kg: 10mg once daily; higher doses of 20mg once daily. Children ≥12 years and Adolescents: 20mg tablet orally once daily.

    No specific antidote for rabeprazole is known. Consult your doctor in case of overdose if unwanted effects occur.
  • Missed Dose

    In case you missed any dose, take the next dose as soon as you remember, but don't double your next dose to compensate for the missed one.

    History of hypersensitivity to rabeprazole or any formulation component, severe hepatic failure, hypomagnesemia.

  • Precautions & Warnings

    Regular monitoring of bone density and calcium supplement is recommended with its chronic use to avoid the risks of bone (hip bone) fracture and with increased susceptibility to certain infections (e.g. hospital- acquired pneumonia, community-acquired clostridium difficile). Rabeprazole should be used with caution in patients with renal impairment or in those at risk of fluid retention. Administration of Rabeprazole to patients with mild to moderate liver impairment resulted in increased exposure and decreased elimination caution should be exercised in those patients.
  • Food Interaction

    Prolonged treatment ≥2 years with Rabeprazole may lead to malabsorption of dietary vitamin B12 and lead to vitamin B12 deficiency.
  • Storage/Disposal

    Store below 25°C in a dry place. Protect from light and moisture.
